目的 探讨柴胡疏肝散对胆囊胆固醇结石(CS)小鼠胆囊收缩功能的影响及其作用机制.方法 将32只ICR小鼠随机分为空白组8只和造模组24只.空白组予以普通饲料喂养,造模组采用高胆固醇致石饲料喂养制备CS小鼠模型,9周后随机选取8只模型小鼠胆囊取材,观察CS造模是否成功.将成模小鼠随机分为模型组和柴胡疏肝散组.柴胡疏肝散组予柴胡疏肝散灌胃,空白组和模型组予等量的生理盐水灌胃,连续干预4周.用ELISA法检测血清胆囊收缩素(CCK)、二酰基甘油(DAG)、甘油三脂(TG)与胆汁总胆固醇(TC)、总胆汁酸(TBA)含量;HE染色法观察胆囊和肝脏的组织形态;分别采用q-PCR法与Western blot法检测肝脏蛋白激酶C(PKC)和胆囊调宁蛋白(CAP)、血管活性肠肽(VIP)基因水平和蛋白表达.结果 与空白组比较,模型组血清DAG、TG及胆汁TC含量明显升高(P<0.05,P<0.01),血清CCK和胆汁TBA含量明显降低(P<0.01),肝脏PKC mRNA与蛋白表达均明显降低(P<0.05,P<0.01),胆囊CAP、VIP的mRNA与蛋白表达均明显升高(P<0.01);与模型组比较,柴胡疏肝散组血清DAG、TG及胆汁TC明显降低(P<0.05),血清CCK和胆汁TBA明显升高(P<0.05,P<0.01),肝脏PKC mRNA表达明显升高(P<0.05),胆囊CAP、VIP mRNA与蛋白表达均显著降低(P<0.05,P<0.01).经药物干预后,柴胡疏肝散组小鼠肝细胞脂肪变性明显改善,胆囊结石状况及组织黏膜胶原纤维改善明显.结论 柴胡疏肝散通过降低DAG、TG及胆汁TC水平并提高血清CCK、胆汁TBA含量,上调肝脏PKC与下调胆囊CAP、VIP蛋白表达,改善胆囊收缩功能,发挥利胆排石的作用.
阿尔茨海默病(AD)已成为全社会迫切关心和亟待解决的健康问题,其病因尚不明确.中西药联用治疗AD的疗效显著,患者认知能力和全身功能得到明显改善,且药物不良反应少.中西药联用可通过多种成分作用于多个靶点而起到全身治疗疾病的作用,更加符合AD的病因复杂、病变部位多样的特点.该文对中西药联用治疗AD的现状进行综述,并对临床上最常用的银杏酮酯-多奈哌齐的治疗疗效进行分析,以期为临床提供参考和借鉴.

Objective The method of intragastric administration of improved guinea pigs was introduced, which can be used as reference for other guinea pigs gavage. Methods 40 ordinary guinea pigs, male and female half, based on the CCKA receptor-mediated G protein signaling pathway in the gallstones of cholesterol gallstones in the study of the mechanism of the establishment of cholesterol gallstones, after the modeling succeed, the use of the stent into the experimental animal esophageal feeding method (traditional gavage method) and the use of the intragastric needle into the experimental animal mouth into the drug by its own swallowing method(improved aftergrowth method), the mortality of guinea pigs after intragastric administration of two different gavage methods was monitored. Results Compared with the traditional gavage method, the mortality of the guinea pigs treated with the improved intragastric method was significantly lower (P <0.01). Conclusion The improved gastric gavage method has high success rate and practicability, so it is worth popularizing.
Objective To explore the features of TCM syndrome elements in polycystic ovary syndrome (PCOS).Methods Standardized to collect four diagnostic data of 100 PCOS patients,analysis of its syndrome elements characterristic by syndrome elements differentiation and statistical analysis.Results ①The main locations of disease in PCOS were liver,uterus,spleen,kidney and skin,liver and uterus syndrome were the most;②The main asthenia syndromes in PCOS was Yin deficiency,the integral of syndrome element of Yin deficiency was significantly higher than that of blood deficiency,Qi deficiency and Yang deficiency respectively (P<0.05);③The main sthenia syndromes in PCOS were heat,Qi stagnation,blood stasis,phlegm,dampness,yang hyperactivity and blood heat.The integral of syndrome element of heat and Qi stagnation were significantly higher than that of blood stasis,phlegm,dampness,yang hyperactivity and blood heat respectively (P<0.01).Conclusion The main locations of disease in PCOS were liver and uterus,hver-qi stagnation was the important pathological mechanism of PCOS.
多囊卵巢综合征是育龄妇女常见的内分泌代谢异常的综合性疾病,其发病机制尚未完全清楚.而从中医学的角度,肝郁是多囊卵巢综合征的重要病因病机之一.运用中医"治未病"思想,以疏肝为要,着眼于情志、运动、针药等中医特色干预手段,对多囊卵巢综合征的防治有重要意义.
Cholelithiasis is a common digestive disease,in which the cholesterol gallstone mainly in recent years,the incidence rate continues to rise.Western medicine treatment mainly for surgical treatment and drug dissolution of the stone,the shortcomings are still faced with residual stones,repeated attacks and other issues.The aim of preventive treatment of disease of TCM is to prevent the disease while treating the disease.It can find the risk factors of stone from the source,and it is of great clinical significance to treat the condition of each stage of the cholelithiasis and to take the corresponding intervention or treatment plan.
We show that hypercholesterolemia contributes to oxidative stress injury progression in brain and simvastatin counteracts the cholesterol-induced peroxidation injury in rabbit hippocampus, and we demonstrate for the first time that the simvastatin is a critical role in brain protection and identify HO-1 and other related antioxidant enzymes as molecular target for active redox compounds. Second, our experiments have pointed out an association between statin treatment and a decrease in the risk of having peroxidation damage of brain. The balance effects of simvastatin to ROS and antioxidants enzymes network are most probably due to improved SOD functional activity, increase in GSH-Px, increase in HO-1 expression, and decrease of MDA generation.
Objective To determine the optimum treatment for viral myocarditis (VMC). Methods A total of 126 VMC patients were randomly divided into the control group (42 cases) that was treated with conventional Western medicine, and the intervention group (84 cases) that was treated with a combination of Chinese medicine (CM) and Western medicine intervention termed optimum proposal of integration of disease and syndrome (OPIDS). Before and after 4 weeks of treatment, the integral of CM syndrome, self-rating depression and anxiety scales (SDS and SAS, respectively), echocardiograms (ECGs), heart rate variability and left ventricular systolic function were observed. Results Compared with the control group, the intervention group showed significant reductions on the SDS and SAS ( P <0.05); improvement of premature ventricular beats, atrioventricular blocks, ST-segment abnormalities, and significant T wave changes ( P <0.05); greater reductions in standard deviation of NN intervals (SDNN), standard deviation for per 5 min averages NN intervals (SDANN), and root-mean-square of successive difference of NN intervals (rMSSD) ( P <0.05); and increases in cardiac output, stroke volume, and ejection fraction, the last of which was statistically significant ( P <0.05). Overall, the treatment efficacy rate was significantly better P <0.05) in the intervention group (75.61%) compared with the control group (69.70%). Conclusion OPIDS is quite effective in treating VMC and improves symptoms such as anxiety and depression, left ventricular systolic dysfunction, premature ventricular contraction, and cardiac autonomic nervous system dysfunction. 目的:研究补肾抗衰片对家兔动脉粥样硬化(AS)病变后血管组织蛋白质硝基化修饰的影响.方法:56只日本大耳白兔随机分为正常组(NOR)、模型组(CHOL)、补肾抗衰片组(BSKS,1 g·kg-1 ·d-1)、辛伐他汀组(SIMVA,5 mg·kg-1·d“);各组分别于给药前、给药后8,12,16周采血,最后1次采血后处死动物,无菌条件下取主动脉,检测血清环氧合酶-2(COX-2)活性以及一氧化氮(NO)、3-硝基酪氨酸(3-NT)水平,检测主动脉诱导型一氧化氮合酶(iNOS) mRNA、p38 MAPK mRNA的表达,p38MAPK阳性面积以及eNOS蛋白水平.结果:与正常组比较,动脉粥样硬化造模的不同时期都可检测到iNOS表达和蛋白质酪氨酸的硝基化,模型组内膜明显增厚、纤维帽较薄,斑块内有大量脂质沉积,血清3-NT水平明显升高,补肾抗衰片干预后,血清3-NT水平下降,NO水平明显升高,而COX-2活性没有明显变化;免疫组化染色发现,主动脉p38 MAPK mRNA,iNOS mRNA基因在正常组仅有少量表达,AS病变组p38 MAPK mRNA,iNOS mRNA表达增加;与模型组比较,补肾抗衰片也明显降低了家兔p38 MAPK水平;p38 MAPK mRNA,iNOS mRNA基因表达明显下降,辛伐他汀也有类似的抑制硝基化效应.结论:动脉粥样硬化时,组织蛋白质酪氨酸发生硝基化损伤,中药复方制剂补肾抗衰片在AS病变中具有重要的抗硝基化作用,其保护机制可能是通过调控p38 MAPK mRNA,iNOS mRNA基因的表达以及影响iNOS/NO-COX-2通路中相关酶的活性,同时补肾抗衰片可能通过对抗硝基化反应稳定动脉粥样硬化斑块.
This study was aimed to determine effect of Bu-Shen Kang-Shuai (BSKS) Tablet on HO-1 mRNA and its associated oxidative stress levels among atherosclerotic rabbits. A total of 56 rabbits were randomly divided into the normal group (8 rabbits) and the experimental group (48 rabbits). Normal diet was given to the normal group. Atherosclerotic rabbits models were established in the experimental group. At the eighth week, rabblits in the experi-mental group were randomly divided into the model group, BSKS Tablet group and simvastatin group. Blood samples were collected before medication, 8-, 12-, 16-week after medication from rabbits of each group. Rabbits were sacri-ficed under aseptic conditions at the last blood collection. Expressions of aortic HO-1 mRNA and PPARα mRNA were measured by Q-PCR method. The level of MMP-9 was measured by immunohistochemical assay. Serum HbCO, COX-2 activity and cGMP level were measured by ELISA assay. The results showed that after the intervention of BSKS Tablet, serum HbCO level decreasd, cGMP was obviously increased. However, there was no obvious change on the COX-2 activity. The immunohistochemical assay showed that BSKS Tablet obviously reduced MMP-9 level of rabbits. There was only small amount of aortic HO-1 mRNA expression in the normal group. However, the expres-sion of aortic HO-1 mRNA in the atherosclerosis group was increased. After intervention of BSKS Tablet, the ex-pression of HO-1 mRNA was increased with statistical significance (P < 0.05). Simvastatin had similar antioxidant effect. It was concluded that the compound preparation of traditional Chinese medicine (TCM) BSKS Tablets had an important antioxidant effect in treatment of atherosclerosis. Its protective mechanism may be through the regulation of HO-1 mRNA gene expression and effects of HO-1/CO-cGMP pathway activities of related enzymes while peroxida-tion stability of atherosclerotic plaque.
Dilated cardiomyopathy is a disease which manifests with heart enlargement, myocardial contractile dys-function and arrhythmia. The disease cause is unknown. And the disease prognosis is not good with high fatality rate. There is no specific treatment for dilated cardiomyopathy in modern medicine. Currently, a large amount of clinical researches have been conducted by traditional Chinese medicine (TCM) scholars with relatively satisfied therapeutic effects. This article made a review on the etiology, pathogenesis, treatment method (including syndrome differentiation and treatment, modern application of ancient prescription, self-made prescription, and Chinese patent medicine liq-uid) and TCM treatment progress of dilated cardiomyopathy in order to show that TCM has certain advantages in the treatment of dilated cardiomyopathy.
There is no specific treatment for the viral myocarditis.With the study of multitude antivirus and immune regulation herbs,the advantages of traditional Chinese medicine are highlighted,making viral myocarditis become one of the diseases for which traditional Chinese medicine has made a breakthrough.Based on the theories of disease and syndrome and corresponding of prescriptions and syndromes,we optimize the strategy of treatment,in order to form composition forces in antivirus and immune regulation and improve the therapeutic effect.
Objectives To evaluate the responsibility of Quality of Life Scale in Patients with Viral Myocarditis (QOL-VMC Scale). Methods Fifty patients who were diagnosed viral myocarditis filled in the QOL-VMC Scale before and after treatment. The responsibility of QOL-VMC Scale was evaluated from both the change of QOL-VMC Scale score and the effect size. Results After treatment the score of the physiological, psychological and social dimensions and the total score of the scale were all higher than before treatment, and the differences were significant (p<0.01). The effect size of the physiological dimension, social dimension and total scale were between 0.52 and 0.65, while the effect size of the psychological dimension was 0.37. Conclusions QOL-VMC Scale is proved with good responsibility, so it can be used to evaluate the therapeutic effect of viral myocarditis.
Atherosclerosis is a condition in which a lipid deposition, thrombus formation, immune cell infiltration, and a chronic inflammatory response, but its systemic study has been hampered by the lack of suitable animal models, especially in herbalism fields. We have tried to perform a perfect animal model that completely replicates the stages of human atherosclerosis. This is the first combined study about the immunologic injury and balloon injury based on the cholesterol diet. In this study, we developed a modified protocol of the white rabbit model that could represent a novel approach to studying human atherosclerosis and vulnerable plaque.
Ethnopharmacological relevance: Si-Miao-Yong-An (Trade name: Mai-Luo-Ning), a Chinese herbal formulation comprising Flos Lonicerae Japonicae, Radix Scrophulariae Ningpoensis, Radix Angelicae Sinensis and Radix Glycyrrhizae Uralensis, has been used in treating ischemic cardiovascular and cerebrovascular diseases for many years. Clinical and experimental studies have shown that Si-Miao-Yong-An can inhibit the inflammatory response and antagonize the blood clotting process.Aim of the study: To investigate the effect of Si-Miao-Yong-An on atherosclerotic plaque stability in rabbit model.Materials and methods: Seventy male rabbits were divided into four groups. Rabbits in the normal group were fed with normal diet, while rabbits in model group and drug treatment groups were fed with high cholesterol diet, underwent BSA-induced immunologic injury and balloon-induced mechanical injury. After atherosclerotic rabbits were treated with simvastatin or Si-Miao-Yong-An for 16 weeks, blood and aorta in four groups were collected for analysis.Results: Si-Miao-Yong-An reduced the level of triglycerides (TG) and low-density lipoprotein cholesterol (LDL-C) in blood after treatment for 16 weeks. Compared with model group, Si-Miao-Yong-An decreased the content of many inflammatory cytokines in blood and plaque. Morphological analysis of abdominal aorta showed that Si-Miao-Yong-An increased fibrous cap thickness and smooth muscle cells, reduced lipid core area and macrophages, and contributed to inhibit matrix degradation and inflammatory response.Conclusion: In this study, we provided evidence for that Si-Miao-Yong-An could promote the stability of atherosclerotic plaque in the rabbit model, indicating that this medicine was a reasonable drug treating cardiovascular diseases in clinical. Objective: To observe the effect of Simiaoyongan Decoction(SYD) on angiogenesis in rabbit's Atherosclerosis(AS) plaque.Methods: 40 Japanese white rabbits were randomly divided in two groups,that is,the control group(n=10,normal diet) and the experiment group(n=30),in which AS vulnerable plaque model was established by feeding high-fat diet,injecting Bovine Serum Albumin(BSA) and balloon catheter overstretching on abdominal aorta artery.At the 8th week,30 animals in experiment group were randomly divided in three groups,that is,model group(n=10),SYD group(n=10),simvastatin group(n=10).The rabbits were sacrificed at the 24th week.Pathological changes were observed by HE,oil red O and Masson staining,and then measured intimae thickness(IT),fiber cat thickness(FCT),FCT/IMT,lipid core area to plaque area ratio(LCA/PA).CD31,VEGF,P38 MAPK protein were detected with immunohistochemistry(IHC),and VEGF mRNA signal was assessed by polymerase chain reaction(PCR).Results: IT in SYD group was lower and FCT,FCT/IMT levels were higher than that of the model group.Compared with the model group,the levels of CD31,VEGF,P38MAPK in the SYD group decreased,and VEGF mRNA expression was inhibited by SYD(P0.01or P0.05).Conclusion: SYD could suppress angiogenesis in AS plaque to stabilize AS vulnerable plaque.It may act by decreasing P38 MAPK,VEGF protein and VEGF mRNA expressions.

OBJECTIVE:To further select the items based on the pre-test version of quality of life scale in patients with viral myocarditis.METHODS:Totally 100 patients with viral myocarditis were enrolled in this study. Methodologies including frequency distribution, discrete trend, t-test, Cronbach's α coefficient, correlation coefficient and factor analysis were applied to select items from different perspectives.RESULTS:A total of 17 items were selected by frequency distribution method from the perspective of central tendency, 15 items were selected by discrete trend method from the perspective of sensitivity, 16 items were selected by t-test method from the perspective of sensitivity and discrimination, 16 items were selected by Cronbach's α coefficient method from the perspective of internal consistency, 12 items were selected by correlation coefficient method from the perspective of representation and independence, and 18 items were selected by factor analysis method from the perspective of representation.CONCLUSION:Item selection of quality of life scale in patients with viral myocarditis was successfully conducted based on the clinical epidemiological data using a variety of statistical methods.
The model of combination of diseases with differentiation of syndromes has a disease name in modern medicine,and adopts treatment principles and methods from traditional Chinese medicine(TCM).It is a new model of combination of TCM and modern medicine.The clinical effect assessment with doctors' experiences of TCM is unable to be accepted internationally.Therefore,it hinders TCM development.However,to adopt standards of modern medicine is unable to reflect advantages of TCM.TCM effect assessment is based on the model of combination of diseases with differentiation of syndromes.It is an integrated method of combining objective effect with subjective effect.This model also combines short term effect with long-term effect.Hence,this model will better reflect the effectiveness and authenticity of TCM.
