目的 探究miRNA-21对2型糖尿病心肌病大鼠心肌纤维化的影响.方法 将32只雄性SD大鼠随机分为四组:对照组、DCM组、DCM+干扰miRNA-21组、DCM+干扰对照组,每组8只.给予DCM组、DCM+干扰miRNA-21组、DCM+干扰对照组大鼠高糖高脂饮食4周后,腹腔注射25 mg/kg链脲佐菌素(STZ),每2周对各组大鼠进行体重、空腹血糖测量,第8周时经尾静脉对DCM+干扰miRNA-21组、DCM+干扰对照组进行慢病毒转染,第12周时所有试验动物处死,进行指标检测;qRT-PCR检测大鼠心肌组织中miRNA-21的表达;HE检测心肌细胞病理学改变;Masson检测心肌组织中胶原纤维的表达;免疫组织化学染色法观察α-SMA的表达.结果 与对照组相比,DCM组大鼠出现体重先增长后下降的趋势,其空腹血糖≥11.1 mmol/L,差异有统计学意义(P<0.05);在转染慢病毒后(注射STZ第8周),与DCM+干扰对照组相比,DCM+干扰miR-NA-21组大鼠体重有所增加,空腹血糖水平下降,差异有统计学意义(P<0.05).与对照组相比,DCM组、DCM+干扰miRNA-21组、DCM+干扰对照组大鼠心肌组织中miRNA-21表达量增加;与DCM+干扰对照组比较,DCM+干扰miRNA-21组心肌中miRNA-21表达量下降,差异有统计学意义(P<0.05);与对照组相比,DCM组、DCM+干扰miRNA-21组、DCM+干扰对照组大鼠出现心肌纤维断裂,周围有炎细胞浸润,且心肌中胶原纤维表达量增加;与DCM+干扰对照组比较,DCM+干扰miRNA-21组改善了心肌纤维的损伤,并降低了胶原纤维表达量,差异有统计学意义(P<0.05);与对照组相比,DCM组、DCM+干扰miRNA-21组、DCM+干扰对照组大鼠心肌组织中α-SMA表达量升高;与DCM+干扰对照组比较,DCM+干扰miRNA-21组的心肌组织中α-SMA表达量下降,差异有统计学意义(P<0.05).结论 miRNA-21参与了DCM的发展过程,抑制miRNA-21的表达可以改善DCM心肌纤维化.
试验以斑马鱼为试验模型,探究虾青素(Astaxanthin)对生长性能及酶活性的影响.将60条斑马鱼幼鱼自由喂养6d再次称重,将所有的斑马鱼分为5组,每组12条.各组分别在基础饲料中添加0(对照组)、0.3%、0.6%、0.9%和1.2%的虾青素.结果 显示,0.6%、0.9%和1.2%虾青素组的斑马鱼增重率(WG)和特定生长率(SGR)均显著低于对照组(P<0.05);各虾青素组的体长增长值均显著低于对照组(P<0.05);各虾青素组血液中酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、溶菌酶(LZM)和超氧化物歧化酶(SOD)活性均低于对照组,各虾青素组丙二醛(MDA)含量均高于对照组.结果 表明,过量虾青素通过降低斑马鱼血液中的酶活性而抑制斑马鱼生长发育..
目的 探讨丹参对异丙肾上腺素(ISO)诱导的心肌损伤内质网应激的干预效果.方法 32只ICR小鼠随机分为4组,即对照组(Con 组),模型组(ISO 组),丹参低、高剂量治疗组(SL 组、SH 组).除 Con 组外,给与各组 ISO,剂量为2mg/(kg·d)腹腔注射4周诱导心肌损伤;第3周开始治疗组同时给与丹参40mg/(kg·d)、80mg/(kg·d)腹腔注射,造模及给药期间Con组和ISO组注射同等剂量生理盐水.实验4周末处死小鼠,称取体质量、心脏质量,计算心脏质量指数(HW/BW);HE和 MASSON 染色观察病理学改变;蛋白印迹法、Real-time PCR 检测 GRP94、IRE1、p-JNK、Caspase-12、Cleaved Caspase-3蛋白及mRNA表达情况.结果 与对照组比较,ISO组心脏质量和HW/BW增高(P<0.05);心肌细胞肥大,纤维化程度增强;GRP94、IRE1、p-JNK、Caspase-12、Cleaved Caspase-3蛋白及mRNA表达明显增加(P<0.001).丹参能降低心脏质量、HW/BW及心肌细胞肥大和纤维化程度,明显降低内质网相关因子表达(P<0.01、0.001),且高剂量治疗效果明显.结论 内质网应激参与ISO诱导的心肌损伤,丹参能够降低内质网相关因子GRP94、IRE1、p-JNK、Caspase-12、Cleaved Caspase-3的表达水平,改善心肌损伤,且高剂量治疗效果显著.
糖尿病是临床上常见的慢性疾病,是一种以高血糖为主要特征的代谢性疾病.糖尿病病因复杂,普遍认为是由于胰岛素分泌缺陷、生物作用受损或由两者共同作用引起,持续的高血糖将会引起心、血管、神经等发生慢性功能障碍或损害,影响患者健康和生活.目前,临床上对于糖尿病以双胍类、INS、格列奈类等降糖药物为主,虽然能降低患者血糖水平,但是长期用药容易增加低血糖、胃肠道不良反应发生率,影响患者健康和生活.桦褐孔菌属于是一种白腐真菌,多存在于亚洲、欧洲及北美洲等,含有多糖成分,具有降血脂、免疫调节、抗氧化等作用,而在糖尿病中的应用研究较少.因此,本文将以糖尿病为起点,分析桦褐孔菌固体发酵菌质水提物在糖尿病中的应用及作用机制,为糖尿病治疗提供依据和参考.
目的:探讨盐酸舍曲林片联合盐酸特拉唑嗪片治疗慢性前列腺炎的疗效.方法:选取2016年6月至2017年9月牡丹江医学院附属红旗医院收治的慢性前列腺炎患者90例作为研究对象,采用系统抽样法将其分为对照组和观察组,每组50例.对照组患者给予维生素C联合盐酸特拉唑嗪片治疗,观察组患者给予盐酸舍曲林片联合盐酸特拉唑嗪片治疗.比较两组患者治疗前后的美国国立卫生研究院慢性前列腺炎症状积分指数、抑郁自评量表评分及临床疗效的差异.结果:治疗后,观察组患者美国国立卫生研究院慢性前列腺炎症状积分指数、抑郁自评量表评分明显优于对照组,差异均有统计学意义(P<0.05);观察组患者的总有效率为93.33%(42/45),明显优于对照组的77.78%(35/45),差异有统计学意义(P<0.05).结论:盐酸舍曲林片联合盐酸特拉唑嗪片治疗慢性前列腺炎的疗效显著.
Objective:To explore the application and effect of IVC independent air supply system in nude mice breeding.Method:In June 2016,a room of about 20 m2was selected from the laboratory animal center of our hospital,on the basis of specific pathogen free(SPF) barrier system,the IVC independent air supply system was installed as the study group;the room of the same size was selected and the SPF barrier system was installed only as the control group.At the beginning of the study,6 female rats (nu/+) and 6 male rats (nu/nu) were placed in the control group and the study group,and the cage was placed according to the ratio of male to female ratio (1︰1).As of December 2017,the delivery interval,average number of litter per litter and survival rate of the two groups were calculated.Result:There were no significant differences between the two groups in the delivery interval and the average litter size per litter(P>0.05).The survival rate of the study group was significantly higher than that of the control group,the difference was statistically significant(P<0.05).Conclusion:The application of IVC independent air supply system in the breeding process of nude mice can significantly improve the survival rate of nude mice,which can provide many high quality experimental animals for clinical animal experiments.
目的:探究在大小鼠生物净化中应用生物工程技术的效果.方法:自2016年1月开始,本研究中心选择常用的C57BL/6J品系小鼠作为研究A组、选择SD大鼠作为研究B组,在繁殖中应用生物工程技术;另选择一部分小鼠与大鼠作为对照A、B组,在其繁殖过程中不做任何干涉随机.每组研究对象15只.实验开始前,对所有鼠的病原微生物进行检测.应用体外受精与胚胎移植的生物工程技术,在HTF培养液中完成受精并发育至2-细胞时期,将胚胎移植入购买来并激素处理过的代孕SPF级ICR雌鼠的输卵管中.代孕ICR雌鼠生下子代,子代3周离乳后,对代孕ICR雌鼠和随机挑选的子代一只进行检测.统计研究A、B组的采集卵子数、2-细胞数与移植胚胎数.统计四组研究对象的平均每胎产仔数、存活率.统计净化前后四组研究对象的病原微生物检出率及净化率.结果:研究A、B组生物工程技术指标(采集卵子数、2-细胞数与移植胚胎数)比较,差异均无统计学意义(P>0.05);四组研究对象平均每胎产仔数与存活率比较,差异均无统计学意义(P>0.05);四组研究对象在合笼与体外受精前,均尾静脉取血进行血霉菌培养.仔鼠出生3周离乳后,每胎随机选一只与其代孕ICR母鼠一起进行尾静脉取血,进行血霉菌培养,对照A、B组的净化率均显著低于研究A、B组(P<0.05).结论:在小鼠大鼠的生物净化中应用体外受精与胚胎移植的生物工程技术,在不影响代孕母鼠的生殖能力与子代存活率的情况下,显著地提升了净化率,达到了研究的目的,从而能为临床动物实验提供更多优质可用的实验动物.
目的:探讨甘精胰岛素联合瑞格列奈治疗2型糖尿病的临床疗效.方法:选取2016年3月至2017年6月牡丹江医学院附属红旗医院收治的2型糖尿病患者86例,以随机数字表法分为观察组和对照组,每组43例.对照组患者给予预混胰岛素治疗,观察组患者给予甘精胰岛素联合瑞格列奈治疗.观察两组患者餐后2h血糖、空腹血糖及糖化血红蛋白水平,临床疗效和不良反应发生情况.结果:治疗后,两组患者餐后2h血糖、空腹血糖和糖化血红蛋白水平较治疗前明显降低,且观察组患者明显低于对照组,差异均有统计学意义(P<0.05);观察组患者的总有效率为93.02%(40/43),明显高于对照组的76.74%(33/43),差异有统计学意义(P<0.05);观察组患者的不良反应发生率为6.98%(3/43),明显低于对照组的23.26%(10/43),差异有统计学意义(P<0.05).结论:甘精胰岛素联合瑞格列奈治疗2型糖尿病的疗效显著,可有效控制患者血糖,且不良反应较少.
Objective To observe the effects of naringin on interstitial fibrosis,signal transducer and activator of transcription (STAT3) phosphorylation in diabetic rats.Methods The male SD rats were administrated with streptozotocin (STZ) by ip to establish diabetic rat model,and then randomly divide into model group,naringin low,middle and high dose (25,50,and 100 mg/kg)group.The normal rats injected with vehicle were designed as control group.Rats in each group were given the corresponding medicine by ig,once a day for eight weeks,while the normal group and the model group were orally administered with saline.At the end of the 8th week in treatment,fasting plasma glucose and heart mass index were measured.Masson staining was used to observe the myocardial fibrosis.qRT-PCR was used to detect the mRNA levels of collagen Ⅰ and fibronetion (FN).Immunohistochemical method was performed to detect the depositions of collagen Ⅰ and FN.Western blotting method was used to detect STAT3 and phosphorylation of STAT3.Results Compared with control group,fasting plasma glucose,heart mass index,the degree of myocardial fibrosis,and the expressions of collagen Ⅰ and FN in left ventricular myocardial tissue of model group were significantly increased (P < 0.05 and 0.01).Compared with model group,fasting plasma glucose,heart mass index,the degree of myocardial fibrosis,and the expressions of collagen Ⅰ and FN in left ventricular myocardial tissue of naringin middle and high dose group were significantly decreased.Conclusion Naringin retards the process of myocardial fibrosis in diabetic rats by downregulating the expression of STAT3,reducing the synthesis and depositions of collagen Ⅰ and FN.
Objective To observe the protective effect of tripterygium glycosides on renal inflammatory injury in diabetic nephropathy rats.Methods 75 male SD rats were used as research materials, including 10 rats in control group and 65 rats in the model group by intraperitoneal injection of streptozotocin diabetic nephropathy rats, and 64 rats were successful.According to the random number table method, the success of extraction the rats were randomly divided into model group, tripterygium glycosides group and rosiglitazone group, and tripterygium glycosides group was divided into 9mg/kg and 18mg/kg.The different groups of rats after treatment on urea nitrogen, serum creatinine, 24h urine protein, serum IL-1β levels, serum IL-17 levels, IFN-y levels were observed and analyzed.Results Comparing with the control group, the blood urea nitrogen, creatinine and 24h urinary protein content, serum IL-1β, serum IL-17 and beta IFN-y levels of the model group increased significantly, and there was statistically significant difference (P<0.05).The 9mg/kg tripterygium glycosides group, 18mg/kg group tripterygium glycosides group and rosiglitazone group was compared with the model group, the urea nitrogen, serum creatinine and 24h urine protein content decreased significantly, and there was statistically significant difference (P<0.05).Comparing with the model group, serum IL-1β, serum IL-17 and IFN-y levels of the tripterygium glycosides group and rosiglitazone group were significantly lower than those of the model group (P<0.05).The difference was statistically significant (P<0.05).Conclusion The tripterygium glycosides can improve renal inflammatory injury in diabetic nephropathy rats, can significantly reduce the level of inflammatory cytokines in serum and kidney, and has a high research value.
Objective To study the protective effect of curcumin on the kidney of type 2 diabetic nephropathy rats.Methods During the period from June 2016 to December 2016, the laboratory 40 male rates were selected and randomly divided into model group and control group, 10 of them as control group with basic diet for a week, only with 30 rats in the model group.The model group was given high fat and sugar on the basis of the basic diet at the same time.The successful model rats in the model group were divided into model group, low dose group and high dose group.The renal pathological changes, blood urea nitrogen, serum creatinine and blood glucose changes in different groups of rats were compared.Results The rats in the control group were administered for 14 days, 21 days, 28 days and 31 days, there was a big difference between the model group on weight, and there was statistical significance (P<0.05).There was a significant difference between the low dose group, high dose group and the model group on weight after administration for 28 days and 31 days, and there was statistical significance (P<0.05).The control group rats and the model group had difference on blood glucose, serum creatinine and blood urea nitrogen levels, and the difference was statistical significance (P<0.05).The high dose group and the model group had significant difference on blood glucose, serum creatinine and urea nitrogen level, and there was statistical significance (P<0.05).The low dose group and the model group had significant difference on serum creatinine urea nitrogen level, and there was statistical significance (P<0.05).Renal lesions in rats of model group, low dose group and high dose group renal lesions were less than those of the model group.Conclusion The curcumin can protect the kidney of type 2 diabetic nephropathy rats.
目的:对实时超声弹性成像技术在梗阻性黄疸引起的肝脏病变中的诊断价值进行研究和分析。方法:选取2014年12月-2017年3月牡丹江医学院附属红旗医院和牡丹江医学院附属第二医院收治的86例梗阻性黄疸患者作为观察组,并按照总胆红素水平的高低将观察组患者具体分为轻、中、重度黄疸三组;同时以同期在牡丹江医学院附属红旗医院和牡丹江医学院附属第二医院接受健康体检的35例志愿者作为对照组。所有研究对象均接受实时超声弹性成像技术的检测,比较相关数据,分析实时超声弹性成像技术在梗阻性黄疸引起的肝脏病变中的诊断价值。结果:观察组患者的MEAN显著低于对照组,AREA%显著高于对照组,差异均有统计学意义(P<0.05)。轻度黄疸组、中度黄疸组、重度黄疸组的MEAN值均明显低于对照组(P<0.05),AREA%值均明显高与对照组(P<0.05);此外,观察组组内两两比较,差异均有统计学意义(P<0.05)。轻度黄疸组、中度黄疸组、重度黄疸组的不同b值时的肝脏ADC值均明显低于对照组(P<0.05);此外,观察组组内,随着黄疸程度的加重,患者肝脏ADC值呈逐渐下降趋势,组间两两间比较,差均有统计学意义(P<0.05)。结论:实时超声弹性成像技术是诊断梗阻性黄疸引起的肝脏病变的有效途径。
Objective To study the mechanism of traditional Chinese medicine Yuye decoction in the treatment of type 2 diabetic rats.Methods During the period from September 2016 to December 2016, 80 rats were chosen as the research objects.All the rats were from the area of animal experimental center of medical research.80 rats were divided into normal group, type 2 diabetes model group, Yuye decoction 4 week group and Yuye decoction 8 week group.The replication model type 2 diabetic rats with high fat and high glucose diet and intraperitoneal injection of streptozotocin, changes of physiological indexes of rats in different groups after 8 weeks were compared.Results The normal group and type 2 diabetes group on FBG, LDL-C, HDL-C, TC, TG, SOD and MDA levels were significantly different, and there was statistical significance (P<0.05).Yuye decoction 4 week group and Yuye decoction 8 week group was compared with type 2 diabetes model group on FBG, LDL-C, HDL-C, TC, TG, SOD and MDA levels, and the difference was statistically significant (P<0.05).Yuye decoction 4 week group and Yuye decoction 8 week group on LDL-C, HDL-C and TG had no statistically significant difference (P>0.05).Yuye decoction 4 week group and Yuye decoction 8 week group on FBG, TC, SOD and MDA levels had statistical significance (P<0.05).The islet volume of the type 2 diabetes model group was significantly smaller than that of the normal group, the number of islet cells of the type 2 diabetes model group was significantly less, the islet volume of Yuye decoction 4 week group and Yuye decoction 8 week group was larger than that of the type 2 diabetes model group, and Yuye decoction 4 week group and Yuye decoction 8 week group had relatively large number of cells.Conclusion Traditional Chinese medicine Yuye decoction can play a role in anti-oxidation, repair of pancreatic beta cells, and can effectively control blood glucose in type 2 diabetic rats.
Objective To investigate the clinical efficacy of tigecycline combined with sulbactam and cefopcrazone in the treatment of lung infection due to multidrug-resistant Acinetobacter baumannii ( MDR-AB ) . Methods Totally 82 patients with lung infection due to MDR-AB admitted to our hospital from September 2015 to September 2016 were divided into two groups according to the random number table method, 41 cases in each group. The control group was treated with sulbactam and cefopcrazone, on the basis of this, the observation group was treated with tigecycline. Results The effective rate of the observation group was 95. 12%, which was significantly higher than 78. 05% of the control group ( P < 0. 05 ) , the bacterial clearance rate of the observation group was 58. 54%, which was significantly higher than 34. 15% of the control group ( P < 0. 05 ) . After treatment, there was no significant difference in leukocyte levels between the two groups ( P > 0. 05 ) , while the C-reactive protein and procalcitonin in the observation group were significantly lower than those in the control group ( P < 0. 05 ) . Conclusion Tigecycline combined with sulbactam and cefopcrazone in the treatment of lung infection due to MDR-AB can obviously improve the inflammatory reaction degree of patients and improve the bacterial clearance rate, and has remarkable clinical curative effect, it is worthy of clinical promotion.
目的 探讨实时超声弹性成像技术(RTE)的弥散定量分析功能对大鼠梗阻性黄疸模型肝脏改变的定量化诊断价值.方法 选取30只2.5~3.0个月龄,体质量280~330 g的雄性SD大鼠,随机分为甲组(实验组)与乙组(对照组),每组15只.甲组大鼠行手术制作可复性梗阻性黄疸动物模型,乙组大鼠仅行假手术.分别于造模前、造模后2周及解除梗阻后2周采用带有组织弥散定量分析软件的超声诊断仪对动物模型进行RTE检查,并对弹性图像进行弥散定量分析;比较分析造模前、造模后2周、解除梗阻后2周两组大鼠%AREA和MEAN值的变化情况.结果 造模前及解除梗阻后2周时两组大鼠的%AREA值及MEAN值比较,差异均无统计学意义(P>0.05);甲组大鼠造模后2周%AREA及MEAN值分别为42.60±3.69、93.20±2.54,与乙组的25.40±1.40、108.00±4.93比较,差异有统计学意义(t=6.230、4.328,P=0.019、0.047);甲组大鼠造模后2周时%AREA和MEAN值分别与造模前及解除梗黄后2周比较,差异均有统计学意义(P<0.01).结论 RTE的弥散定量分析功能可反映梗黄引起的肝脏改变,梗黄后肝脏ROI区MEAN及%AREA可有显著改变,其可作为梗阻性黄疸程度的定量化评定参考指标.
Objective To explore the research progress of curcumin in treating diabetic nephropathy. Methods Literature datas of cur-cumin in the treatment of diabetic nephropathy were retrieved in the VIP,Wanfang,CNKI,Cochrane library,et al. To investigate the related characteristics of diabetic nephropathy,summarize the main pathogenic factors and the value of curcumin in the treatment of dia-betic nephropathy. Results Diabetic nephropathy was a common disease and needed to be paid attention,the treatment combined with using curcumin can effectively improve the clinical effect,improve blood glucose,stabilize the disease,and had high medical value. Conclusion In the treatment of diabetic nephropathy,effective methods should be adopted to control the patient′s condition. Curcumin has the effects of regulating blood lipids,anticoagulation,antioxidation,bile,anticancer and so on,it can greatly improve the condition of the patient with diabetic nephropathy,which is worthy of clinical promotion.
1 大肠杆菌病 大肠杆菌病是由致病性大肠杆菌所致的人畜共患病.病型复杂多样,主要引起腹泻、败血症,各器官局部感染及中毒症状等.败血型表现发热,精神不振,常见于出生7天以内的犊牛.病犊体温升高到41℃以上,精神萎顿,卧地不起,腹泻,排出水样稀便,呈淡黄色,常混有血块、血丝和气泡,具恶臭味,多于发病后1~2内死亡,从血液或内脏易于分离到致病性血清型大肠杆菌;肠毒血型由特异血清型大肠杆菌增值产生肠毒素吸收后引起,没有菌血症,较少见,常突然死亡,多见于生后1周龄内的犊牛,病程稍长的呈中毒性神经症状,先兴奋后沉郁,较少见腹泻,最后昏迷死亡;肠型见于1周龄后的犊牛,病初体温升高到39.5~40℃,粪便开始为黄色,后变为灰白色,混有凝乳块、血丝或气泡,有酸败气味,病后期,大便失禁,体温正常或下降,脱水而死亡.病程长的可出现肺炎、关节炎、脐炎,患犊牛痊愈后发育迟缓.
目的是通过壳聚糖对血糖、血脂、胰岛素的影响探讨壳聚糖对糖尿病的作用机制,有效降低血糖和血清胆固醇浓度,提高胰岛素的相对生物利用率,壳聚糖在体内直接转染胰岛素基因治疗糖尿病,有望成为治疗Ⅰ型糖尿病的胰岛素基因载体。
