目的:体外研究琼脂糖用于肺腺癌细胞株A549三维培养,并观察细胞在三维环境中的行为改变.方法:用琼脂糖作为三维培养基质,观察细胞在三维环境中的生长现象;黏附实验用以检测细胞对胞外基质的黏附率;MTr比色法检测细胞对DDP的药物敏感性和细胞增殖抑制率;流式细胞术检测细胞自然凋亡情况.同时比较二维及三维培养细胞的差异.结果:在琼脂糖中,A549细胞生长呈现球形的团块状生长,细胞团块大小不一,生长状态良好.随着细胞的增长,3D培养的A549细胞对DDP的抗药性增加,药敏性降低;二维和三维培养下的A549细胞黏附率存在显著差异;3D培养下的细胞自然凋亡明显减少.结论:A549细胞在琼脂糖三维培养中,其细胞行为与二维培养时发生显著改变,二者具有显著性的差异.因此,琼脂糖可用于构建A549三维培养模型,此模型可为进一步研究提供研究基础.
The aim of this study is to investigate the mechanisms of Chinese herbal medicine called “Baoyuan Jiedu” (BYJD for short) decoction, improving life quality and preventing muscle atrophy of cancer cachexia model mice. We showed that the effect of BYJD decoction increased body weights of mice and reduced tumor volume and tumor mass. Furthermore, BYJD decoction increased the gastrocnemii mass and the transverse diameter of muscle fiber morphology. Moreover, BYJD reduced the expression of Atrogin-1 and MuRF-1 protein. Collectively, our results show that BYJD decoction improves the life quality of cancer cachexia mice and prevents muscle atrophy by downregulating expression of Atrogin-1 and MuRF-1.
保元解毒汤是基于肿瘤发病病机是“阴寒内盛,邪毒凝滞”的癌毒思路指导下形成的.当前对该方的研究主要集中在本方干预癌性恶病制的药效及机制研究方面,为了更全面地了解复方的化学组成概况,进一步探讨复方干预肿瘤微环境的药效机制,指导分析分离组方的效应物质基础,文章将各组成药的化学成分进行了系统地整理和概述,同时基于复方中不同药物组合功效,从组成药物对肿瘤微环境中免疫及炎性机制的影响,进行梳理和评价.
Objective To observe the characteristics of liver-kidney yin deficiency syndrome in late stage of spontaneously hypertension rats (SHRs). Methods Ten 12-month-old healthy male SHRs were put into syndrome observer group (SHR group), while ten male WKY rats with the same age were set as control group (WKY group). Syndrome differentiation method on rats was employed to transform the four-diagnostic information into general behavior indexes with the same significance. The afternoon anal temperature, tongue color, holding power, ability of learning and memory, serum estradiol and testosterone level were observed, compared and analyzed. Results Compared with the WKY group, rats in the SHR group had higher afternoon anal temperature, redder tongues, poorer strength and claw holding power and their ability of learning and memory was retarded; the level of serum estradiol was lower;the ratio of estradiol to testosterone was higher. All of these were largely in line with deficiency of live-kidney yin syndrome. Conclusion SHRs in late stage show characteristics of liver-kidney yin deficiency syndrome.
目的 探讨肺腺癌A549细胞三维立体(3D)培养模型对顺铂(DDP)的耐药性及其黏附耐药机制.方法 胶原培养条件下建立A549三维立体培养模型,MTT法检测平面(2D)培养和3D培养A549细胞敏感性.采用整合素β1激动剂(TSF)和抑制剂(MAB)干预,对其相关机制进行研究.结果 A549细胞3D培养模型中可见细胞生长良好,细胞之间黏附形成典型的多细胞球.2D培养模型中A549细胞对DDP的IC50值为12.84 μg/ml;3D培养A549细胞对DDP的IC50是31.74 μg/ml,为2D培养的2.47倍.与空白对照组比较,DDP干预组及MAB+DDP组的抑制率明显升高(P<0.01),TSF+DDP组有较显著差别(P<0.05);与DDP干预组相比较,TSF+DDP组细胞抑制率明显降低(P<0.05),MAB+DDP组明显升高(P<0.05).结论 胶原法成功建立A549细胞的3D培养模型.与2D培养相比,该细胞模型具有耐药的特性,其机制可能与促进整合素β1对细胞的黏附作用有关.
通过考辨小肠清浊功能发展源流,认为《内经》、《难经》中文字差异为分歧之源头,易水学派将小肠功能归属于胃,中西汇通医家依据西医解读小肠,泌别清浊功能逐渐边缘。重新梳理小肠泌别清浊功能,有利于继承《内经》理论,加深对唐宋方书相关文献的理解,丰富发展中医藏象理论。
目的 了解血瘀体质女性在正常女性中的比率,分析女性血瘀体质的影响因素. 方法 2013年7月至11月,在山东省济南市采用随机抽样的方法开展流行病学调查,采用中医体质分类与判定表测定研究对象体质,采用自制女性血瘀体质影响因素调查表及总体幸福感量表测定血瘀体质的影响因素.结果 女性血瘀体质与职业,学历,饮食的喜成与喜辛辣,运动规律,久坐,睡眠方面的睡眠规律、质量、失眠、多梦,流产史,疾病史中的心脏病、良性肿瘤、月经不调、痛经、子宫疾病、妇科炎症以及紧张焦虑与抑郁等情绪状况有显著相关性(P<0.05);饮食的喜咸与喜辛辣、睡眠规律、失眠、运动规律、流产史、良性肿瘤、月经不调、妇科炎症、紧张焦虑、抑郁是女性血瘀体质的重要预测因子.结论 喜食咸味与喜食辛辣、熬夜或不规律的睡眠、失眠、缺乏运动、流产、良性肿瘤、月经不调、妇科炎症以及紧张焦虑、抑郁等因素对女性血瘀体质的形成具有一定的影响.
Objective: To summarize the modern theoretical basis for the treatment of tumor by Wen Yang method and its connotation,and discuss the significance of Wen Yang method to treat tumor and classify the research of experiment and clinic about treating tumor by Wen Yang method. Methords: A Total of 206 articles about treating tumor by Wen Yang method published between 2003 ~ 2013 were retrieved in CNKI with the key words of warming the Yang,tumor,clinical research,experimental study. Inclusion criteria: 1) Study on the theory and connotation of Wen Yang method for treating tumor; 2) Experimental study of antitumor effect of Wen Yang herb extraction monomer; 3) Experimental study on Wen Yang method therapy for tumor 4) Clinical study on Wen Yang method therapy for tumor. Forty papers were collected. Results: Studies of Wen Yang method for tumors are more concentrated on the experience from the old TCM doctors,Wen Yang herb extraction monomer,compound anti tumor,etc. Lack randomized controlled studies of large scale systems. Conclusion:Wen Yang method for treating tumor has a great significance,especially combined with western medicine therapy. The best treatment for tumor is the choice of treatment by traditional Chinese and western medicine,particularly giving the first place to TCM treatment with Wen Yang method is one of the important measures for integrated Chinese and western medicine therapy for the prevention and treatment of tumor.
目的:研究西黄滴丸对S180肉瘤小鼠的抑瘤作用,并探讨西黄滴丸对荷瘤小鼠肉瘤组织中血管生成标记物(CD34)和相关因子血管内皮生长因子(VEGF)表达的影响.方法:采用S180肉瘤小鼠模型,分为模型组,西黄丸组(2.2 g·kg-1·d-1),西黄滴丸低、中、高剂量组(1.5,3,6 g·kg-1·d-1)以及环磷酰胺组(CTX,0.02 g·kg-1),西黄丸组,西黄滴丸低、中、高剂量组ig给药,环磷酰胺组ig给药,每2d1次,连续10 d给药;观察西黄滴丸对S180荷瘤小鼠的抑瘤作用,并采用HE染色观察肿瘤细胞的形态变化,免疫组织化学的方法观察西黄滴丸对荷瘤小鼠肿瘤CD34及VEGF蛋白表达的影响.结果:与模型组比较,滴丸高剂量组荷瘤小鼠体重有明显升高;西黄丸组,西黄滴丸低、中、高剂量组及CTX组抑瘤率分别是26.28%,21.47%,26.92%,30.76%,35.85%;CD34的蛋白表达CTX组较模型组有明显降低(P<0.01),西黄滴丸高、中、低剂量组明显降低CD34蛋白表达(P<0.05),滴丸高剂量组较西黄丸组有较明显差异(P<0.05);滴丸中、高剂量组及CTX组与模型组比较VEGF的蛋白表达有明显降低(P<0.05),滴丸中剂量组及CTX组较西黄丸组蛋白表达有较明显差异.结论:西黄滴丸对S180荷瘤小鼠具有抑瘤的作用,其机制可能与下调肿瘤组织CD34及VEGF蛋白表达水平有关.
目的 分析女性血瘀体质的特征,旨在为女性血瘀体质的辨识提供新的理论依据.方法 于2013年7月至2013年11月采用《中医体质分类与判定表》的血瘀体质分量表、自制女性体质特征调查表、艾森克人格问卷(成人版)、总体幸福感量表对济南市女性开展临床流行病学调查.结果 血瘀体质女性和平和体质女性在冷环境适应能力、常见症状(皮肤状况、牙龈状况、脱发状况、睡眠状况、月经状况)有显著性差异(P<0.05),人格特征方面,二者在艾森克人格问卷的E、N、P、L分量表均有显著性差异(P<0.05),心理特点方面,二者在总体幸福感量表的总量表、精力、对生活的满足和兴趣、忧郁或愉快的心境、对情感和行为的控制、松弛与紧张(焦虑)方面均有显著性差异(P<0.05).结论 血瘀体质女性在环境适应能力、常见症状、人格和心理方面都有较为独特的特征.
To explore the apoptosis mechanism of Wenxia Changfu Formula (ae (c) aie, e...ae-(1), WCF) in reversing drug resistance of lung cancer in vivo.Thirty model mice were randomly assigned to three groups: control group, cisplatin (CDDP) group, and WCF group. A transplanted tumor model of lung adenocarcinoma was established in all groups. Mice in the WCF group received intragastric administration of WCF (0.2 mL/10 g body weight) everyday in addition to CDDP intraperitoneally (5 mg/kg body weight) twice a week. The mice in the CDDP group received CDDP intraperitoneally (5 mg/kg body weight) twice a week, while the control group received normal saline intraperitoneally (0.2 mL/10 g body weight) everyday. The weight of the nude mice and respective tumors, tumor volume and tumor-inhibiting rate were measured. Electron microscopy was used to observe the existence of apoptosis body. Apoptosis index (AI) was detected by TdT-mediated dUTP nick end labeling staining. The expression of Fas and FasL mRNA was investigated by reverse transcription polymerase chain reaction, while immunohistochemistry was applied to detect the protein expression of Fas and FasL, caspase-3 and caspase-activated DNase (CAD), respectively.Compared with CDDP group and control group, WCF could significantly reduce the tumor volume from the 19th day and alleviate the tumor weight (P < 0.05), and the apoptosis body was found in tumor cells in the WCF group. WCF could also enhance the level of AI, up-regulate the expression of caspase apoptosis pathway related protein caspase-3 and CAD, as well as the expression of Fas, FasL mRNA and protein (P < 0.05).WCF could improve the sensitivity of tumor cells to CDDP and reverse the drug resistance by inducing the apoptosis.
Cultivating and propagating socialist core values should be based on outstanding traditional culture. Shandong University of Traditional Chinese Medicine attaches great importance to cultural features of Chinese medicine to combine practicing socialist core values together with propagating culture of traditional Chinese medicine. TCM classroom teaching becomes an important resource to spread contents of socialist core values. Campus culture construction is carried out to demonstrate characteristics of Chinese medicine.
目的 研究西黄滴丸对S180荷瘤小鼠的抑瘤作用及其机理.方法 将S180荷瘤小鼠分为模型对照组,西黄丸组,西黄滴丸低、中、高剂量组及环磷酰胺(CTX)组.称取小鼠体重及瘤重,计算抑瘤率;并采用免疫组织化学的方法检测西黄滴丸对S180荷瘤小鼠肿瘤组织中PCNA及CyclinD1表达的影响.结果 西黄丸组,滴丸低、中、高剂量组及CTX组抑瘤率分别是26.28%,21.47%,26.92%,30.76%,35.85%.与模型对照组相比,滴丸高剂量组和CTX组瘤重有显著性降低(P<0.01),滴丸高剂量组PCNA光密度值具有明显差异(P<0.05).与模型组相比,滴丸中剂量组及CTX组Cy-clinD1光密度值具有显著性差异(P<0.01).结论 西黄滴丸对S180荷瘤小鼠具有抗肿瘤的作用,其机理可能与下调肿瘤组织中PCNA及CylinD1的蛋白表达有关.
目的 通过文献研究探讨高血压病肝肾阴虚证的症状特征,提炼核心症状.方法 检索中国知网学术期刊全文数据库涉及高血压病肝肾阴虚证的文献及其文献中非肝肾阴虚证的症状、证候、诊断研究内容等,对符合标准文献中肝肾阴虚证和非肝肾阴虚证的症状进行出现频次和频率比较分析,并筛选组间差异显著症状,经logistic回归分析,建立预测模型,ROC曲线评价预测效果.结果 共纳入符合标准文献104篇,涉及证型症状描述301条,其中肝肾阴虚证证型104条,非肝肾阴虚证证型197条.经比较筛选得到18个症状纳入回归模型,其中有意义症状5个,按回归系数由高到低排序依次为:腰膝酸软、脉弦细、阴虚发热、舌红少苔、健忘,均为正向影响肝肾阴虚证的诊断因素.ROC曲线评价诊断价值较高.结论 在高血压诊断明确的基础上,腰膝酸软、脉弦细、阴虚发热、舌红少苔、健忘为高血压病肝肾阴虚证的核心症状.
目的 检测不同加工莱阳沙参的氨基酸含量.方法 采用氨基酸自动分析仪对不同加工的莱阳沙参进行氨基酸检测.结果 莱阳沙参至少含有19种氨基酸,不同的加工品氨基酸含量差异明显,烫去皮晒干北沙参总氨基酸含量最高(19.18%).结论 为不同加工的北沙参质量评价提供依据.
The Shandong University of Traditional Chinese Medicine has taken cultivating talents with culture as soul engineering in moral composition and talent rearing. TCM culture, modern science culture and self-culture of the university were brought into the system of moral composition and talent rearing. The system takes TCM cultural courses as core, TCM cultural research as key, old-generation experts’scholarship and experience as basis, integration of Qilu culture and TCM culture as feature and brand feature cultural activities as carrier for constructing the system of cultivating talents with culture and improving moral composition and talent rearing.
围绕临床医学专业学位研究生课程教学的主要环节和影响因素进行调研,较为系统的了解我国目前临床医学专业学位研究生课程教学现状,发现在课程设置、教学方法等方面存在的问题.
Shiji Yaoyu is the existed and the earliest work entirely explaining TCM nursing among TCM literatures. It is also the first work on nursing in Chinese history. Qian Xiang in Qing Dynasty wrote Shiji Yaoyu,and there are existed three versions of it. The academic thought of this book originated from Neijing and had the collection of the ideas of different schools. Regarding the academic thought,Qian Xiang stressed on the nursing of emotions,life nursing of patients and the observation of the sick condition of patients,as well as the normalization of medicinal nursing. Even though there were shortcomings in Shiji Yaoyu,it was still the good example for the study on TCM nursing before the Qing Dynasty,which plays the important role in TCM nursing history. The academic thought has profound impacts and brings the essential reference value to the modern TCM nursing.
采用文献学研究方法,系统梳理《诸病源候论》中小肠藏象理论,认为小肠具有运化水液,主司小便,与三焦膀胱共同形成机体津液代谢通道,功能失常则小便涩少,可致痰饮、消渴等;心与小肠相表里,小肠助心通利水饮,心火可下移小肠,心与小肠主血,与冲任共主妇人经带.
