’Longfan No.14’ is a new middle-late maturing tomato F 1 hybrid developed by crossing inbred line ’13117’ as female parent and ’13118’ as male parent.It is of indeterminate growth type.Its plant has strong growing vigor.The first inflorescence sets on the 7-9th segments,with inflorescence intervals of 3-5.The fruit is of round shape and red in color.Its young fruit has no green shoulder.The average single fruit weight is 170.5 g.The fruit hardness is 2.09 kg·cm -2 ,abnormal cracked fruit rate is 2.89%.The soluble solid content is 5.12%,VC content is 90.2 mg·kg -1 ,and organic acid content is 3.9 g·kg -1 .It is highly resistant to TMV,resistant to TYLCV and leaf mold,tolerant to early blight.It can yield about 105 t·hm -2 .It is suitable to be cultivated in protected conditions in spring at Gansu Province.
[背景]在全球气候变化的背景下,大气CO2浓度的升高会影响植物病害的发生,进而影响农业生产.β型碳酸酐酶(β-carbonic anhydrase,βCA)是植物CO2感应和浓缩系统中的重要组成元件,参与拟南芥和烟草的植物免疫过程,但在番茄(Solanum lycopersicum)等园艺作物中的研究较少.[目的]通过探究番茄S1βCA3在抵御植物病害中的作用及机制,为番茄生产中的抗性调控提供科学依据.[方法]以拟南芥AtβCA氨基酸系列为参考序列,在番茄So1 genomics network数据库中鉴定到4个S1βCA.进一步以野生型(wild-type,WT)番茄'Ailsa Craig'(AC)为材料接种丁香假单胞菌番茄致病变种(Pseudomonas syringae pv.toma to DC3000,Pst DC3000),利用 qRT-PCR 技术测定叶片中S1βCA的表达量,筛选出受Pst DC3000诱导表达的基因S1βCA3.在此基础上,以AC为背景,利用农杆菌介导法进行番茄遗传转化,构建S1βCA3稳定过表达植株(OE-S1βCA3).通过观察OE-S1βCA3植株接种PstDC3000后的抗性表型,明确S1βCA3在番茄抵御Pst DC3000过程中的作用.为了研究S1βCA3调控植物抗病性的内在机制,比较WT和OE-S1βCA3植株接种PstDC3000与对照条件下转录组的变化,并利用KEGG数据库对差异基因进行功能分析,推测糖代谢与S1βCA3介导的免疫反应有关.最后,通过测定WT和OE-S1βCA3植株糖代谢及其信号途径相关基因表达量以及葡萄糖、果糖和蔗糖含量,对转录组结果进行验证及分析.[结果]OE-S1βCA3植株对PstDC3000的抗性增强,接种Pst DC3000后,叶片中的细菌生长量、病斑数以及死细胞积累量明显减少.转录组测序结果显示,正常条件下,OE-S1βCA3植株转录谱没有发生明显变化;接种PstDC3000后,在WT和OE-S1βCA3植株中检测到2 100个PstDC3000诱导基因,其中有63.3%的基因在OE-S1βCA3植株中表达量更高.KEGG分析结果显示,依赖于S1βCA3过表达的Pst DC3000诱导基因富集在糖代谢相关路径中,包括淀粉和蔗糖代谢,内质网中的蛋白质加工(糖基化),氨基糖和核苷酸糖代谢,真核生物中的核糖体生物合成以及光合作用等路径.糖代谢与糖信号密不可分,qRT-PCR及糖含量测定结果显示,接种PstDC3000后,OE-S1βCA3植株叶片中糖代谢及其信号传导途径相关基因表达量与葡萄糖、果糖和蔗糖的含量较WT更高.[结论]番茄S1βCA3的过表达增强了植株对Pst DC3000的抗性,该过程可能与糖代谢及其信号通路在植物免疫中的作用有关.
以青海湖裸鲤(Gynnocypris przewalskii)为研究对象,采用半定量和定量PCR法研究HCO3-分泌相关基因SLC4(solute carrier family 4)和SLC26(solute carrier family 26)家族slc4a1、slc4a2、slc4a4和slc26a6基因组织分布情况,并对不同盐碱环境下肠道中SLC基因家族的表达情况进行定量分析,揭示青海湖裸鲤适应盐碱环境的肠道调节机制.结果表明,slc4a1、slc4a2、slc4a4和slc26a6基因在青海湖裸鲤鳃、肝脏、肾脏和肠道等多个组织中均有表达,且在肠道中的表达量较高,其中slc26a6在肠道中高表达,而在鳃中表达量极低,表现出组织特异性;在碱度组[盐度1.31 ±0.02,碳酸盐碱度(30.66±0.08) mmol·L-1]、盐度组[盐度15.02±0.02,碳酸盐碱度(2.12 ±0.05) mmol· L-1]和湖水组[盐度14.84 ±0.03,碳酸盐碱度(29.57±0.11) mmol·L-1]青海湖裸鲤肠slc4al、slc4a2、slc4a4和slc26a6基因的表达量在胁迫4d过程中均呈现出先升高后回落的现象,其中湖水组裸鲤肠SLC4、SLC26家族基因表达量上调最为明显,尤其是slc26a6基因表达量最高上升为对照组的4.9倍,同时,湖水组裸鲤直肠排泄HCO3-浓度也最高,说明盐碱环境下青海湖裸鲤通过肠道Cl/HCO3-交换子(slc4a1、slc4a2、slc26a6)、Na+-HCO3联合转运子(slc4a4)分泌和排泄机体内积累的碱,这一调节途径有助于青海湖裸鲤补偿因水环境中盐碱度升高而造成的渗透及酸碱失衡.

开展了几种硝酸盐调控剂种类、组合、使用浓度、使用时期对西芹硝酸盐含量影响的研究,试验结果表明:在西芹采收前使用双氰胺、硫酸锰、钼酸铵、硫酸锌、赤霉素等硝酸盐调控剂,均可降低西芹的硝酸盐含量,其中以双氰胺、硫酸锰效果佳;双氰胺与硫酸锰、钼酸铵、硫酸锌、赤霉素的组合对西芹硝酸盐积累的抑制效果优于单施双氰胺,可使西芹硝酸盐含量接近WHO/FAO规定的限量标准。钼酸铵最佳使用浓度为600mg/L,硫酸锰最佳使用浓度为0.05%,硫酸锌最佳使用浓度为0.15%,赤霉素使用浓度可控制在25~50mg/kg。从使用时期来看,采收前10d为最佳使用时期。
Puionium Gaertn,a kind of sand vegetable mostly for its leaf.Several nutrient contents of leaves in different plantlet were investigated,evaluating its dynamics,providing academic for the first harvesting date.The result showed that: the contents ofvitamin C,protein,amino acidand soluble sugar of had a 8~12 leaves in P.cornutum(L.)Gaertn and P.dolabratum Maxim were high.At the same time,plant were in the growth peak period,so leaves of P.cornutum(L.)Gaertn and P.dolabratum Maxim could be harvested for the first time.
The effects of low temperature were investigated on the content of N and chlorophyll in pepper leaves under controlled conditions of sunshine used greenhouse in this paper.The results showed low temperature decreased the content of N.The treatment of 15 ℃/9 ℃ reduced significantly the content of N,which decreased of 6.63 % after 15 d compared with 0 d.Both 15 ℃/9 ℃ and 10 ℃/5 ℃ treatments for 15 d reduced the content of Chl a,Chl b and Chl(a+b).The treatment of 10 ℃/5 ℃ decreased 45.6 % of Chl a,42.5 % of Chl b and 4.48 % of Chl(a+b) compared with the treatment of 28 ℃/18 ℃.There was a positive relationship between the nitrogen content and chlorophyll content.
The experiment was conducted with seeds and seedlings of sweet pepper,treated by different exogenous hormones,to probe into effection to growth and senescence in sweet pepper.The results showed,treated by S3307,IBA+NAA,6-BA and GA3,they significantly increased leaves number and leaf area Index.Meaniwhile,they increased plantlet yield and staved drop speed of chlorophyll and soluble protein content except GA3.In addtion,they also increased dry matter weight and MDA content and improved activity of SOD and POD treated by 4 exogenous hormones.
The flow mathematical model of the crude oil is described with the relaxation theory of elastomer and the reptation theory of the viscous body.The impacting of some correlated fators on the low-temperature flowing of the crude oil is discussed.The flow characteristic of the crude oil has relationship not only with the temperature,but also with the shear stress,shear time,shear modulus and viscocity of the crude oil.Among them,temperature has the most significant influence.A method of improving the low-temperature flow characteristic of the oil and correlated modifier is provided.
Influence of Ca(NO3)2 on permeability of plastic membrane,MAD content,the activity of protective enzyme and substance of protective content were studied under 5 ℃ using pepper seedling as materials.The results showed that Ca(NO3)2 treatment increased significantly the activity of protective enzyme and substance of protective content,decreased electrolytic leakage and MAD content,improved the ability of resistance at low temperature.The concentration at 10 mmol/L showed the best.
对5种植物生长调节剂防止辣椒落花及其增产效应进行了研究。结果表明:在辣椒植株开花期间,用纯天然植物生长调节剂爱密廷40000倍液、50000倍液及NAA 50mg/L液喷花,其座果率、每100个果重及产量均比对照清水喷花或其它处理显著增加,其中用爱密廷50000倍液喷洒,辣椒座果率为86.5%,较对照提高17.7%;平均每100个果重2.41kg,较对照提高了10.6%;产量3673.33kg/m2,较对照增加28.7%;苯酚类植物生长调节剂2,4-D喷洒植株,虽然也能提高辣椒座果率,但容易产生药害。

Effects of different concentrations of Ca2+ and inhibitors of Ca2+ signal system on some osmolytes,including soluble protein,soluble sugar and proline in pepper under low temperature stress were investigated.The results indicated that extraneous calcium(10mmol/L) treatment could markedly increase the content of soluble protein,soluble sugar and proline.Moreover,addition of Ca2+ chelating agent EGTA or calmodulin antagonist W7 could markedly decrease the content of soluble protein,soluble sugar and proline.Taken together the above results,it is suggested that Ca2+ signal system may be regulated by change of some osmolytes in Pepper under low temperature.
The effect of seeds soaking with Ca2+ and calmodulin antagonist W7 on chilling resistance of pepper seedlings were studied.Seed soaking with Ca2+ solution obviously decreased the contents of malondialdehyde(MDA) and electrolyte leakage,increased the activities of superoxid dismutase(SOD),the peroxide enzyme(POD) and the content of soluble protein and soluble sugar of pepper leaves,seed soaking with W7 distinctly enhanced the contents of MDA and electrolyte leakage,reduced the the activities of SOD,POD and the content of soluble protein and soluble sugar of pepper seedling leaves under chilling stress.The result suggests that Ca2+·CaM messenger system plays an important role in controlling chilling resistance of pepper seedling.
This research was about the effeceion of low temperate and weak light on two hot pepper varietis.The results were as follows:in the situation of 100-2μmol·m-2·s-1,23℃/10℃,the concentration of chlorophyl-a and chlorophy-b in hot pepper leaves increased,while the ratio of chlorophyl-a /chlorophy-b reduced;according to the reduction of temperature and light the concentration of chlorophyl-a and chlorophy-b went down,while the ratio of chlorophyl-a /chlorophy-b went up;after the treatment of low temperature and weak light,concentration of carotenoids reduced in hot pepper leaves.
研究了不同定植方式和密度对日光温室辣椒生长和产量的影响,结果表明,单株定植和双株定植对辣椒叶片叶绿素含量及地上部、地下部干重无明显影响,随着单株定植密度的增加,地上部干重呈下降趋势;单株定植较双株定植可显著提高辣椒根系伤流量和粗根数;定植密度为83370株/km2时,单株定植(株距20 cm)的前期产量和总产量分别为22816 kg/km2、98 227 kg/km2,较双株定植(对照)分别增加13.7%和11.2%,宜在生产中推广使用。
